No. The manual calculator only needs you to know roughly which direction each room is in. If you do have a floor plan image, the Floor Plan tool lets you tag rooms visually on it instead.
It is as accurate as your compass readings. For a quick check it is very reliable for the big rooms — kitchen, master bedroom, entrance, pooja room. For borderline cases near a direction boundary, the floor plan tool is more precise.
